Many diseases arise from genetic variation. Genetic variants play a central role in determining disease onset and progression not only in rare genetic disorders caused by single-gene mutations, but also in a broad range of diseases, including cancer and neurodegenerative disorders. Approaches to studying and treating these diseases can be broadly divided into two directions: understanding the functional consequences of disease-associated genetic variants and directly correcting the causal variants. However, the same genetic variant can produce markedly different functional outcomes depending on the cell type, developmental stage, and physiological context. Therefore, both the accurate interpretation of variant function and the effective application of genome-editing technologies require a thorough understanding of cell type-specific biology.
From this perspective, our laboratory is establishing a large-scale platform for functional variant analysis using human pluripotent stem cells and precise genome editing technologies. This platform enables the systematic characterization of genetic variants across different cell types and developmental stages. We also investigate the interplay between cell type-specific biology and genome editing and integrate genome editing technologies with artificial intelligence to develop innovative genome editing platforms. Ultimately, by advancing technologies for both understanding and correcting genetic variants, we aim to elucidate the molecular mechanisms of disease and establish foundational technologies for next-generation gene therapy and precision medicine. -
